A New Photobioreactor for Continuous Microalgal Production in Hatcheries Based on External-Loop Airlift and Swirling Flow ArchiMer

The combination of the state-of-art with the constraints inherent to hatcheries has turned the design into a closed, artificially illuminated and external-loop airlift configuration based on a succession of elementary modules, each one being composed of two transparent vertical interconnected columns. Relationships exist between environmental factors, physiological state of oysters, defence system and pathogens. Hatchery oysters are more and more produced and reared in the field. The French oyster farming industry relies almost exclusively on juveniles collected from the natural environment; the supply of spat produced by hatcheries is low, about 10% of the industry's requirements. Development through selective breeding of oyster stocks which are better suited for aquaculture purposes, is likely to reverse this tendency since only hatcheries will be able to supply such animals. Comparison of growth, digestive system maturation and skeletal development in sea bass larvae reared in an intensive or a mesocosm system ArchiMer

The quality of development in intensive or mesocosm hatchery-reared Dicentrarchus labrax larvae was investigated using physiological indicators assessing ontogeny. Larvae were reared in intensive (120 L tanks) and in mesocosm systems (20 m(3) enclosures) with the same feeding sequence, excluding the wild zooplankton naturally available in mesocosms. Faster growth was recorded since early development [16 day after hatching (DAH)] in the mesocosm. This thesis describes the conception and the study of a photobioreactor dedicated to aquaculture. The state of the art and the technico-economic constraints proper to this activity led to the choices to a simple reactor, volume variable, cheap to produce and reliable for continuous cultures. The elementary module is made of two vertical columns, connected by flanges. In order to study the genetic influence of intensive production of the larval stage in commercial hatcheries, two types of specific selective processes were examined: the effects of discarding the smallest larvae and temperature. A mixed-family approach was used in order to infer the genetic composition of the larvae. The results show that our mixed-family approach, combined with microsatellite-based family assignment, is a powerful tool for the study of bivalve larvae genetics. Selective sieving is an advantageous practice at a phenotypic scale, but also represents a substantial risk for diversity loss. Pacific oyster Crassostrea gigas is one major species for aquaculture, and the development of breeding programs and the need for preservation of wild stock genetic resources prompted the need for larvae cryopreservation. The objective of the present study was to choose the most reliable protocol from several existing publications, to test its biological and practical limitations, and to adapt it to hatchery conditions. The selected protocol was characterized by a very slow freezing rate without seeding, and by the use of ethylene glycol and sucrose as cryoprotectant. The aim of this project was to evaluate the impact of probiotic (Phaeobacter gallaeciensis, X34 strain) treatment on the complete development (from veliger to metamorphosis) of Pecten maximus larvae in the context of a bacterial challenge and in conditions more representative of hatchery practices. Establishment of microbiota in larval culture of Pacific oyster, Crassostrea gigas ArchiMer

This study has two main objectives: (1) to implement a recycling aquaculture system (RS) for the larvae of the oyster Crassostrea gigas, and (2) to characterise the bacterial communities established in different compartments of this system. An RS with 25% fresh seawater addition per hour and another with no addition (0%) were compared with a flow-through system (FT). Larval survival was equivalent in RS and FT, but growth rate was 17% slower in RS than in FT. The physical chemical parameters remained stable, except for pH that decreased to 7.75 and salinity that increased to 37.5‰ in the RS 0%. Although batches of oyster and clam larvae have been routinely reared in the hatchery of Argenton without antibiotics, high larval mortalities have been recorded with Pecten maxirnus under similar conditions.
